
Effect of Fetal Bovine Serum Concentration on Lysophosphatidylcholine-mediated Proliferation and Apoptosis of Human Aortic Smooth Muscle Cells.
Lysophosphatidylcholine (lysoPtdCho) produced by phospholipase A2-mediated hydrolysis of phosphatidylcholine and can stimulate the proliferation and apoptosis of smooth muscle cells of blood vessels. We investigated the effect of bovine serum (FBS) concentration of the fetus in the culture medium at lysoPtdChomediated apoptosis and proliferation of human aortic smooth muscle cells (HASMCs) as well as the activation of extracellular signal-regulated kinase (ERK) 1/2.
In the presence of 1% FBS, HASMC increased survival after treatment lysoPtdCho at 1 and 10 pM but decreased at 25 and 50 pM. However, the increased survival lysoPtdCho HASMC dose-dependent manner in the presence of 10% FBS. Caspase 3/7 activity in HASMCs increased by 25 pM lysoPtdCho in the presence of 1% FBS, but not 10% FBS.
Furthermore, lysoPtdCho at 1 and 10 pM induced ERK1 / 2 phosphorylation in the presence of 1% FBS, but not in 10% FBS. Thus, lysoPtdCho HASMC-mediated apoptosis, proliferation, and ERK1 / 2 activation depends on the concentration of FBS.
Fetal bovine serum attenuation-inducing perfluorooctanoic acid toxicity to human cell lines some through albumin binding.
perfluorooctanoic acid (PFOA), a typical emerging organic pollutants, can interact with serum albumin. However, still characterize the binding of PFOA in serum albumin and to address the role of these interactions related toxic effects. We aimed to characterize the interaction between PFOA and albumin to understand the effects of these interactions on the absorption, distribution, and toxicity of PFOA in human cells.
The results showed that the number of viable cells was significantly enhanced by the addition of fetabl bovine serum (FBS) into the cell culture medium with 300 pM treatment PFOA. PFOA is mainly existed as a complex with FBS, the FBS concentration> 10%, which substantially reduces the absorption efficiency of all cell lines PFOA.
The majority of PFOA accumulates in the cytosol fraction, followed by a nucleus, and mitochondria. Conclusively, our research shows that organic contaminants complexation with protein may reduce their toxicity by reducing cellular uptake.

High quality screening fetal bovine serum for pig oocyte maturation in vitro.
Fetal bovine serum (FBS) is widely used in cell culture due to the high stability and easy access. It is also used as a substitute pig follicular fluid (PFF) in previous studies.
However, components of FBS is not clear, and the FBS in the culture medium can introduce variations from batch to batch. This study aims to establish an effective method to screen in place PFF FBS in culture media for pig oocytes in vitro. We screened FBS from different sources using fibroblast cells from fetal pigs.
Effects of six FBS samples on the growth of fetal pig fibroblast cells were tested through test frozen cell survival, cell clone formation assay, cell growth curve, and the parts of the cell test activities. Serum best we called GFBS (heat-inactivated FBS, cat 10500-64; .. Gibco) showed similar effects on pig oocyte maturation and development with the PFF and can be used as a good substitute for PFF.
